Description
The The invention relates to devices for the treatment of suspended biological Particles with electric fields, in particular electrode chambers for the electroporation and / or electrofusion of biological cells or cell constituents, electrode supports for such devices, and Method for treating suspended biological particles with electric fields, in particular electroporation and electrofusion processes.
Neil, Eds. CRC Press, London, 1996 Electromanipulation of Cells). The Treatment of biological cells or cell components with electrical Fields for modifying the structure and / or material composition the treated particles are well known (see U. Zimmermann, G.A. Neil, Eds. CRC Press, London, 1996 Electromanipulation of Cells). In electroporation, the membrane of biological cells under Effect of electric fields reversibly reversible for a short time, allowing substances from a suspension fluid into the cell interior can be channeled. In electrofusion, two (or more) cells or cell components become lodged Effect of an electric field merged with each other, tasks the electrofusion exist, for example, in the production of hybrid cells, the genetic material of both fusion partners in a common cell nucleus contain, or from modified cells, where the fusion products from cells of one fusion partner and cell components of the second Merger partners. The latter application is particular in the modification of antigen-carrying cells (eg dendritic cells) Cells) with constituents of tumor cells of interest.
Überhöhte Suspensionsdichten ergeben nicht-reproduzierbare Porations- oder Fusionsergebnisse.conventional Chambers for electroporation or fusion of biological particles exist from a suspension container to take on a cell suspension and several arranged in the suspension tank Electrodes. The electrodes are electrically connected to a control device connected to application-dependent be subjected to certain voltages or voltage sequences. The electrodes are shaped and arranged so that in the suspension liquid inhomogeneous electric fields for electrical treatment of the cells be generated. Experiences with the following explained Types of conventional Porous or fusion chambers have shown that the treatment outcomes in particular of geometric properties of the suspension container and Depend on the electrode assembly. It is particularly necessary that the suspension density between the Electrodes are not too high, otherwise, for example, in electroporation act on the particles different strong electric fields or In the case of electrofusion uncontrollable multiple fusions occur. Excessive suspension densities yield non-reproducible Poration or fusion results.
At high suspension densities, although a reversible permeabilization (electroporation) is possible. However, there is the problem that, for example, suspended cells are exposed to different field strengths depending on their position relative to the electrodes. Since electroporation is sensitively dependent on field strength based on the integrated Laplace equation, there is a variation in the degree of permeabilization of the cells and thus in the transfection rates. For the most uniform possible uptake of foreign molecules into the permeabilized cells (small range of variation), the cell density in the suspension should be less than about 10 6 cells / ml.

A common problem with conventional pore and fusion chambers is that the cell suspensions are typically provided at relatively high cell densities (eg, about 10 9 cells / mL), but poration or fusion is provided at low densities in the range of 10 5 to 10 10 6 cells / ml should be performed. Although it is possible to adjust the optimal cell density by dilution. However, this results in large volumes of liquid, the conventional chambers are not suitable for their treatment, since they can not be easily enlarged. Greater electrode spacings would require higher voltages for poration or fusion and a reduction in the conductivity of the suspension liquid. The ready The provision of higher voltages (in the range of, for example, 10 kV and above) would be too expensive and too difficult for practical, in particular mass applications in medicine.

The The object of the invention is improved devices and methods to provide electrical treatment of biological particles, with which the disadvantages of conventional Techniques overcome and in particular the treatment of larger cell suspension volumes under defined and reproducible field conditions with a allow high yield. The invention is also intended to facilitate the practical use of poration and simplify fusion procedures in medicine. Devices according to the invention should also be for designed a simplified observability of the treatment outcome be.
These Tasks are with an electrode chamber and a method with the features according to claims 1 and 6 solved. Advantageous embodiments and applications of the invention will be apparent from the dependent claims.
The The basic idea of the invention is, in the case of an electrode chamber, in particular for the treatment of suspended biological cells or cell components, at least one electrode carrier Provide with at least one pair of electrodes, wherein the electrode carrier a Carrier body with has at least one planar side surface on which the at least a pair of electrodes is arranged. The arrangement of at least a pair of electrodes that in particular for the production of Porations- or Fusion fields is set up on the planar side surface of the electrode carrier has the advantage that, on the one hand realizes the favorable field conditions can be as they are known from the helical chamber, and on the other hand a systematic treatment of large suspension volumes with high yield allows becomes. With the at least one planar side surface of the electrode carrier is defines a reference plane. Adjacent to the reference plane becomes a Half space formed, from which the particles to be treated under the effect of negative or positive dielectrophoresis to the electrodes or can be attracted. In this half-space, with suitable control of the electrodes an electroporation treatment is performed. An electrofusion treatment however, it is transversal within the reference plane or parallel for aligning the respective substantially planar side surface of the carrier body.
According to one preferred embodiment of Invention are in a suspension tank several compartments formed, each having at least one electrode carrier with associated with at least one planar aligned electrode pair is. The compartments can through the body or the walls of the suspension tank are formed, as it is known, for example. Of titer plates. In this Case is preferably in each compartment at least one electrode carrier to the Floor and / or deck areas arranged the compartment. It is also possible that the compartments through the electrode carrier to be self-educated. The suspension container is, for example, a tub for receiving the cell suspension into which several electrode carrier used as inner part walls are. This embodiment the invention is particularly suitable for the treatment of large volumes of suspension, the individual electrode carrier preferably be driven sequentially, as explained below.
An electrode carrier according to the invention preferably consists of a plate-shaped, at least one-sided, substantially planar carrier body with two side surfaces. Depending on the application, the side surfaces are referred to as the upper and lower side surfaces in the horizontal operating position of the electrode carrier and as the front or rear side surfaces in the vertical operating position of the electrode carrier. At least one of the side surfaces carries at least one pair of electrodes arranged in two electrically separate from each other th electrodes. The electrodes are connected to a control device and can be subjected to a predetermined voltage or to perform a fusion treatment relative to each other with predetermined fusion voltages to form certain field conditions in the half-space adjacent to the side surface. The electrodes are shaped such that inhomogeneous electric fields are formed between the electrodes of an electrode pair. The inhomogeneity is given when the electric field strength in the region between the adjacent electrodes is location-dependent. In order to produce the inhomogeneity, a shaping or structuring of the electrodes and / or the side surfaces of the electrode carrier is preferably carried out. The electrodes form electrically conductive strips that protrude from the side surface of the electrode carrier. They are formed, for example, by fixed wires or thin or thick layers. The possibly predetermined structuring of the electrode carrier comprises outstanding portions on which the electrodes are arranged.
One inventive method to treat biological particles with electric fields characterized in that a suspension of the particles to be treated, in particular the biological cells or cell components, introduced into a suspension container in which a plurality of electrode carriers each having at least attached to a pair of electrodes on at least one side surface is, and a treatment of the particles between the electrode carriers takes place. It is preferably a pre-treatment (eg electroporation) of Particles between the electrode carriers and a subsequent dielectrophoretic collection the particle on at least one side surface between the electrodes the at least one electrode pair provided. Then an electrofusion takes place by dielectrophoretic alignment of particle chains between the electrode pairs and subsequent exercise of at least one fusion pulse. The collection of the particles and the electrofusion become at electrode chambers with several compartments preferably sequentially in the individual Compartments performed sequentially in time.
The inventive method is preferably carried out with the electrode chamber according to the invention. It but it is also possible in particular the said sequential treatment of one in several Compartments subdivided suspension with curved electrode carriers or usual electrode arrays perform.
According to one advantageous design of the invention, at least one electrode carrier with the aligned particles while maintaining the current field conditions from the suspension tank taken and transferred to a monitoring and measuring device (eg microscope) become. The Invention has the following further advantages.
